Summary: Experiments conducted in this thesis were developed taking into account its practical implementation, so that we could hear, more profound, applying the technique SCSA for determining the status of sperm chromatin in different species, setting the grade maturation of chromatin at different levels epididimarios (head, body and tail) and the ejaculate in three species (ram, dog and deer), as well as seasonal variations in the same (ram, deer and brown bear). Moreover, the ability to estimate in bull fertility through rates of no return has enabled us to establish correlations between the quality of sperm chromatin (SCSA both as a kit Halomax in its two variants) and fertility, thus confirming the importance of the application of these techniques. Lastly, we have demonstrated the importance of morphometric measurements of the head sperm automated by its correlation with the quality of chromatin in eyaculados of brown bears. We can summarize our research on sperm chromatin in four main areas: comparison between different sections epididimarios and eyaculados study seasonal relation to fertility and relationship with the sperm morphometry. As for the study of the maturation epididimaria we have concluded that there are different points in the sperm chromatin condensation along the epididymis depending on the kind (of body glue in sheep and head to body and red deer dog). In addition, samples eyaculadas and from the tail of the epididymis are at high degree of condensation of the chromatin in the three species, which means that they could be used in the creation of gene banks. With respect to seasonal study, in sheep, there is a greater descondensación of chromatin during the summer months (July and August), which could estarinfluida by the effect of high temperatures. In the case of deer and bear, sperm chromatin appears more descondensada during the reproductive period, which could be related to higher rapjidez passage through the epididymis. As for the study of condensation of the chromatin in bull SCSA determined by DNA fragmentation and determined by Sperm-Bos -Halomax are highly correlated with fertility, can differentiate between groups of fertility very próximales, the percentage of acrososmas intact in the total population, the standard deviation of DFI (detected by SCSA) and the percentage of DNA framgentado (detected with the test Sperm-Bos-Halomax in fluorescence microscopy). The condensation of the chromatin epsermática in brown bear correlates with the morphometry of the sperm head determined by automated analysis. However, both techniques appear to explain various aspects of the variability among males. In addition, elproceso cryopreservation in this species, reduces the size of the sperm head.
